Understanding CNS Depression and Its Impact on Mental Health

Another major cause of CNS depression in people who have no history of using CNS depressants is brain injury. If your doctor suspects that a brain injury or tumor is the cause of your depression, they might order a CT scan, an MRI scan, or both. II. Part 2. Stimulants and Depressants

In the SCN lesion + ANA-12 group, the mRNA levels and circadian rhythm amplitude of BDNF were markedly lower than those in the SCN lesion group (Fig. 6a–c). Similarly, the protein expression levels of TrkB, BDNF, p-ERK/ERK, and p-CREB/CREB were significantly decreased in comparison to the SCN lesion group (Fig. 6d–h). In contrast, no significant differences in these protein expression levels were observed between the sham and sham + ANA-12 groups (Fig. 6d–h). Unlike other forms of depression that primarily impact mood, CNS depression affects most bodily functions, including breathing, heart rate, and blood circulation. They not only reduce anxiety and induce sleep but also slow down essential bodily functions, which can be dangerous in high doses. Central Nervous System (CNS) depression is a condition where the brain and spinal cord’s activity slows down, leading to reduced alertness, drowsiness, and even life-threatening symptoms in severe cases.

Interestingly, uncontrolled neuroinflammation may represent a common pathological substrate for further neurological diseases other than MS and psychiatric disorders. In this context, increased production of TNF-α and decreased levels of the anti-inflammatory mediator transforming growth factor-β (TGF-β) have been considered as neurobiological links between depression and AD 208,209. Multiple brain circuits dysfunctions are thought to contribute to the pathophysiology of psychiatric symptoms in CNS disorders, and to regulate their manifestations. These networks can be investigated by means of techniques measuring neuronal oscillations (i.e., electroencephalogram, EEG; magnetoencephalography, MEG) and brain imaging 72,73,74,75,76,77,78,79,80,81,82,83. For this reason, in this review we will focus on neuroimaging correlates of brain networks alterations. BDNF plays an important role in the pathogenesis of anxiety and depression 30.

Apathy may have a prognostic value in all of the considered neurodegenerative diseases, since it seems to predict a faster cognitive decline 5,25,55,59. In MS, depressive symptoms occur with a prevalence of 50% or higher during the years that immediately follow the diagnosis 7,60,61. The onset of depression may be vague and involve clinical features that are frequently misinterpreted as typical MS symptoms, namely fatigue and sleep, appetite loss, memory and concentration disturbances 22. The possible under-diagnosis of depressive symptoms in MS may contribute to the reduction of health-related quality of life, and to the occurrence of further complications, such as suicidal ideation 30. Psychiatric and neurological diseases often overlap with each other and co-occur 1.

Circadian rhythms significantly influence human mood fluctuations, as established in prior studies 5, 33. This research elucidates how dysfunctions in the suprachiasmatic nucleus (SCN), the master regulator of circadian rhythms, impact mood disorders such as anxiety and depression. Our findings highlight the critical role of the BDNF-TrkB pathway in the striatum, influenced by SCN dysfunction, in the manifestation of these disorders. In this study, we developed a mouse model with SCN lesions and observed disturbances in the 24-h circadian regulation of core body temperature, along with behaviors indicative of anxiety and depression.

For the regional measures of each node, the critical values for a two-tailed test, maintaining a type I error of 0.01, were set as the 99th percentile points of each distribution. The effect sizes (Cohen’s d) for the differences in network measures between the two groups were calculated to quantify the magnitude of group differences. Furthermore, to investigate the association between network measures and illness severity, Pearson partial correlation analysis (controlling for age and sex) was conducted between network metrics and BDI-II scores for the ScD group.

More recently, different investigations focused on blood p-tau as a more easily accessible biomarker reflecting AD pathophysiology. Preliminary findings indicated that blood p-tau shows a stepwise elevation along the AD continuum, being increased even in the preclinical stages, with respect to healthy controls and other neurodegenerative disorders 214. Thus, high-performance assays targeting blood p-tau could represent the first simple, practical, and non-invasive test to rule out AD in case of pure psychiatric debut. The effects of depression can interfere with a person’s ability to manage relationships, work, and daily activities, such as eating and sleeping. Healthcare professionals also refer to the condition as major depressive disorder (MDD) and clinical depression. Some researchers believe that staging mental health conditions like depression can protect against a one-size-fits-all approach that may result in under or over treating some individuals.

As a recreational drug, people sometimes call them barbs, downers, or phennies, among other names. In 2020, the Food and Drug Administration (FDA) strengthened their warning that benzodiazepine use can lead to addiction. Combined with alcohol, opiods, and other CNS depressants, they can be life-threatening.
